Surface coupling of oligo-functionalized dendrimers to detect DNA mutations after blocked isothermal amplification

This study presents a useful strategy for the bioconjugation of probes developing high-performance versatile chips applied to fast-response, low-cost DNA biosensing. We herein demonstrate that high-density immobilization dendrimer-oligonucleotide hybrids promotes reliable sensitive sensing single-nucleotide variants despite their low concentration. Carboxyl-terminated poly(amidoamine) dendrimers directly coupled amine-derivatized oligonucleotides were anchored activated surfaces thermoplastics (polycarbonate and cycloolefin polymer). Surface characterization techniques hybridization assays reported multiple functional sites oligo-functionalized dendrimer facilitated efficient capture targets (5 pM detection limit). Array performance was better than functionalized with linear crosslinkers, such as vinyltriethoxysilane or 3-(triethoxysilyl)propyl isocyanate, well films unconjugated molecules. Based on oligo-dendrimers hybrids, disposable DNA-based biosensing platforms developed point-of-care diagnostics. A selective high-sensitive assay performed included amplification products blocked PCR recombinase polymerase (RPA) an isothermal reaction. The chip results indicated mutant variant BRAF oncogene correctly discriminated in colorectal tissues from cancer patients.
